As shown in the trailer, Butcher is looking to make the Supe problem go away, albeit differently. With his terminal illness from using temporary Compound V and Victoria Neuman getting closer to being the POTUS, Butcher is turning to quick and fast measures. Viewers who have seen the Gen V will remember the Supe virus which only targets the Supes. By looks of it, the trailer also references it indirectly and it seems the Butcher's got his eyes on a bio-engineered virus.
Homelander has a new aim in this season 4 of The Boys. He is targeting "corruption" though whether this "corruption" is in Vought or the government of United States is unclear from the trailer. He also has changed the Seven's aim from being a superhero squad to a group of wrathful gods.
All in all, the new season's looking to amp up the diabolical factor.

The Boys Season 5 has been picked up for season 5. The showrunner Eric Kripke, who was the showrunner for Supernatural may also be looking to wrap the series within 2 more seasons. Speaking at the Amazon's unfront where renewal for The Boys season 5 was announced, Kripke spoke about The Boys being the best job he'll ever have and his only problem was what to write for season 5 since this year promised to be free of conflict or misinformation.
